Friability Test Procedure — Regulatory Guidelines:-
The friability test is a pharmaceutical quality-control test used to determine the resistance of uncoated tablets to abrasion, chipping, and mechanical shock during handling, packaging, and transportation.
The procedure is generally based on the applicable pharmacopoeia, such as USP–NF General Chapter (Tablet Friability), the Indian Pharmacopoeia (IP), or the European Pharmacopoeia (Ph. Eur.). Your approved product specification and applicable pharmacopoeial edition take precedence.
Apparatus and requirements:-
•Friability tester (commonly Roche-type).
•Calibrated analytical balance.
•Tablet dedusting brush or other suitable de dusting methods.
•Representative sample of tablets.
Controlled laboratory conditions as specified in the approved method.
STEP BY STEP PROCEDURE:-
The following is the general procedure for compressed, uncoated tablets based on USP–NF General Chapter
1. Sample selection
- Tablet unit weight ≤ 650 mg: take whole tablets totaling approximately 6.5 g.
- Tablet unit weight > 650 mg: take 10 whole tablets.
2. Dedusting: Carefully remove loose dust from all tablets before weighing.
Initial weighing (W₁): Accurately weigh the dedusted tablet sample and record the initial weight.
3.Initial weighing (W₁): Accurately weigh the dedusted tablet sample and record the initial weight.
4. Load the apparatus: Place the tablets inside the friability drum and close it securely.
5. Set the operating conditions: Rotate the drum for 100 revolutions at 24–26 rpm under the current USP–NF procedure.
6. Collect the sample: Remove the tablets carefully and de-dust them using the same method as before testing.
7. Final weighing (W₂): Accurately weigh the tablets after the test and record the final weight.
8. Inspect the tablets: Check for cracks, cleavage, broken tablets, and other visible damage.
CALCULATIONS OF FRIABILITY:-
Example
•Initial weight (W₁) 6.500 g
•Final weight (W₂) 6.450 g
•Weight loss 0.050 g
•Friability 0.769%
ACCEPTANCE CRITERIA:-
Test parameter & General requirement
Friability :- NMT 1.0% weight loss for most conventional tablets
Visible damage:- No obviously cracked, cleaved, or broken tablets
Repeat testing:- If results are difficult to interpret or exceed the target, repeat twice and calculate the mean of three determinations
Important USP SPECIFICATIONS:-
Parameter & USP requirement
•Test :- Friability of compressed, uncoated tablets
•Drum internal diameter:- 283–291 mm
•Drum depth:- 36–40 mm
•Rotation speed:- 25 ± 1 rpm
•Number of revolutions:- 100
•Sample for tablets ≤650 mg:- Approximately 6.5 g total
•Sample for tablets >650 mg:- 10 whole tablets
•Acceptance limit :- Generally NMT 1.0% weight loss
Important clarification: Under the revised USP text, a weight loss of not more than 1.0% is considered acceptable for most products for a single determination or the mean of three determinations. Follow the product-specific specification and current official USP text.
The USP–NF chapter considers weight loss of not more than 1.0% acceptable for most products. Effervescent, chewable, and other special tablets may require different limits. Follow the registered product specification where it differs.

EXPIRED MEDICINES💊:MYTH vs REALITY😱
Many people assume that once a medicine reaches its expiry date ,it instantly becomes toxic or completely ineffective. The reality is more nuanced.
MYTHS❌:
Expired medicines become poisonous the day after the expiry date.
REALITY ✅:
Most medicines don't suddenly turn toxic after the expiry date. The expiry date indicates the period during which the manufacturer guarantees the medicine's full potency, safety and quality when stored correctly.
MYTHS ❌:
Expired medicines work just as well forever.
REALITY ✅:
Overtime many medicines gradually lose their effectiveness .While some may retain much of their potency beyond the expiry date ,others can degrade significantly, leading to reduced therapeutic benefits .
IMPORTANT CONSIDERATION 📌:
•Potency may decrease after expiry.
•Storage conditions (heat, humidity , sunlight) affect stability.
•Certain medications are more sensitive to degradation than others.
•Expired medicines should not be relied upon for the critical condition where accurate dosing is essential.
THE TAKEAWAY 📌:
Expiry dates are not arbitrary - they are based on scientific stability testing. While expired medicines are not necessarily dangerous, using in-date medicines remain the safest and most reliable choice to ensure optimal treatment outcomes.
As a healthcare professional, educating patients on the proper use ,storage and disposal of medicine is just as important as prescribing them.
